Electrophysiology is the measurement of current or voltage from electrically active cells within living organisms, from muscle cells to neurons. This measurement can occur at a single-cell or ion channel level, or be the result of simultaneous activity in hundreds or thousands of cells. Electrophysiological techniques provide a unique insight into the workings of ion channels and systems within living organisms. Ultimately, this research can be applied to human studies, leading to improved diagnosis and treatment – giving patients better outcomes. To measure low amplitude cellular currents or voltages, electrophysiologists require highly sensitive equipment to provide precise and reliable control in a range of applications – instruments include electrophysiology system manipulators or micromanipulators and a wide selection of complementary accessories.Why use electrophysiology system manipulators?
Electrophysiologists use electrophysiology system manipulators or micromanipulators to move very small instruments or tools in experiments conducted under microscopic visualisation. While these-highly trained professionals have exceptional fine motor skills, using purpose-designed devices to complete these delicate positioning and movement tasks is much more effective, as it removes the limitations which pure manual control would impose. Each micromanipulator translates macroscopic movements of the human hand into microscopic movements of the tools, micropipettes and microelectrodes, critically needed for successful electrophysiological recordings..Functions and applications of electrophysiology system manipulators
Micromanipulators and their accessories allow electrophysiologists to complete a variety of experiments and tasks, including but not limited to:- Making electrical recordings with micropipettes from single-living cells or preparations, such as brain slices
- Manipulating electrical stimulation electrodes
- Holding perfusion pipettes for the localised application of pharmacological compounds
- Dissecting tissue or single-cell structures
- Completing acute or chronic stereotaxic experiments
The types of electrophysiology system manipulators
Electrophysiology system manipulators generally use three different drive mechanisms – which control their movements. These are either mechanical, hydraulic, or motorised/electronic, each having advantages or disadvantages in certain settings. As well as selecting an appropriate drive mechanism for a micromanipulator, scientists will need to decide between the capabilities of each electrophysiology manipulator. Typically the most important features include:- The type of movement – coarse or fine movement control, or both
- The direction of movement – be that in single and three-axis manipulators
- The movement range - typically manipulators with fine movement have more restricted overall ranges
Mechanical
Mechanical micromanipulators use mechanical gears or cantilevers to reduce macroscopic movement into microscopic ones. It should be noted that mechanical drive mechanisms may not be suited to projects that include higher magnification as the micromanipulator can vibrate the instrument when dials are turned.Motorised or electronic
Motor-driven micromanipulators also use similar gears to mechanical models but are driven by electrical motors that are controlled by remote buttons or joysticks, reducing the risk of vibration.Hydraulic
Hydraulic micromanipulators may incorporate some mechanical elements but for the most part, use the movement of hydraulic fluid and like their motorised equivalents allow for remote control. Hydraulic manipulators are completely free of electrical interference, which can be a consideration in some applications. Macroscopic movements are scaled down to a microscopic level as the fluid is transferred between cylinders or pistons of larger or smaller diameters. If you need an electrophysiology system manipulator requiring improved instrument stability – as in patch-clamp recordings with motionless micropipettes – consider a water hydraulic model.
